Autoconfocal microscopy with nonlinear transmitted light detection

چکیده
We describe a simple and robust technique for transmission confocal laser scanning microscopy wherein the detection pinhole is replaced by a thin second-harmonic generation crystal. The advantage of this technique is that self-aligned confocality is achieved without the need for signal descanning. We derive the point-spread function of our instrument and quantify both signal degradation and background rejection when imaging deep within a turbid slab. As an example, we consider a slab whose index of refraction fluctuations exhibit Gaussian statistics. Our model is corroborated by experiment. © 2004 Optical Society of America OCIS codes: 110.0180, 180.1790, 180.5810, 030.6600, 190.4160.

Autoconfocal microscopy with a cw laser and thermionic detection.
We introduce an application of thermionic emission in a PMT photocathode. Because of the nonlinear dependence of thermionic emission on absorbed laser power, a conventional PMT is found to produce a virtual pinhole effect that rejects unfocused light at least as strongly as a physical pinhole. This virtual pinhole effect is exploited in a scanning transmission confocal microscope equipped with ...
متن کاملAutoconfocal transmission microscopy based on two-photon-induced photocurrent of Si photodiodes.
We describe a simple, self-aligned confocal transmission microscopy technique based on two-photon-induced photocurrents of silicon photodiodes. Silicon detectors produce photocurrents in quadratic dependence on incident intensity under the pulsed illumination of light with wavelengths longer than 1.2 microm. We exploit this nonlinear process to reject out-of-focus background and perform depth-s...
متن کاملEnhanced fluorescence signal in nonlinear microscopy through supplementary fiber-optic light collection.
Nonlinear microscopy techniques crucially rely on efficient signal detection. Here, we present a ring of large-core optical fibers for epi-collection of fluorescence photons that are not transmitted through the objective and thus normally wasted. Theoretical treatments indicated that such a supplementary fiber-optic light collection system (SUFICS) can provide an up to 4-fold signal gain. In ty...
متن کاملStaining with two observational methods for the diagnosis of tuberculous meningitis
Ziehl-Neelsen (Z-N) staining of cerebrospinal fluid (CSF) for acid-fast bacilli (AFB) is the cornerstone of the laboratory diagnosis of tuberculous meningitis (TBM). However, the sensitivity of conventional Z-N staining for the detection of AFB in CSF specimens is suboptimal. The present study aimed to compare the practicality of modified Z-N staining with light microscopy and fluorescence micr...
متن کاملLow-energy STEM of multilayers and dopant profiles.
A conventional scanning electron microscope equipped with a LaB6 source has been modified to operate in a scanning transmission mode. Two detection strategies have been considered, one based on the direct collection of transmitted electrons, the other on the collection of secondary electrons resulting from the conversion of the transmitted ones. Two types of specimens have been mainly investiga...
